Biorad CFX96 Real-Time PCR

The Biorad CFX96 instrument is mainly used for accurate quantification of sequencing libraries, before running them on our sequencing systems. The sequencing libraries are quantified using the KAPA Library quantification kit, which is sold by the Core facility as a service. The Core Facility is also equipped with a Qubit 4, which quantifies sequencing libraries by measuring fluorescence from dsDNA binding dyes. However, KAPA library quantification is more accurate, as it is based on primers binding to specific Illumina adapter sequences in the DNA fragments. Thus, only quantifying sequenceable DNA fragments, while the Qubit quantifies total dsDNA in the library prep.
