Vilber Newton 7.0 FT500

The Newton 7.0 offers bioluminescence and fluorescence imaging studies.

More information about the In Vivo Imaging System can be found here.

Location

The imaging system is located at 16-2-8 (lab id 227 056) inside the 16-2 quarantine animal unit (IVC unit at 16.2) in a GMO2 room with rack for housing and safety benches for working. Access rule are the same as for the rest of 16-2 and you need a valid permit to work with animals and a permit from Department of Experimental Medicine, AEM, before you can start working here.
On your first visit to 16.2 please introduce yourself to the animal caretakers. They like to know who is doing IVIS experiments, and if any special precautions should be taken.

Excitation filters

  • 11-position motorized, preinstalled filters: 440nm, 480nm, 540nm, 640nm, 680nm, 740nm, 780nm (BW: 40-50nm)

Emission filters

  • 500nm, 550nm, 600nm, 650nm, 700nm, 750nm, 800nm, 850nm (BW: 20-50nm)

F.O.V.

  • 6x6 cm to 20x20 cm

Software

  • License-free, we have a copy you can get, but also check with the company for upgrades.

Tissue transmission

Tissue is not very transparent in the blue and green region, where stains such as DAPI and GFP emit light. Shifting to the red and especially near infrared area greatly increases signal transmission.

Tissue auto fluorescence

Mouse tissue has a certain degree of auto fluorescence, strongest in the 500-600 nm region and weakest in the >800 nm region.

Chow auto fluorescence

Normal chow contains chlorophyll which emits light in the 700 nm region. This is especially critical when imaging in the stomach and gut area. Therefore, a tracer in the above 800 nm region should be chosen. Also, the chow could be changed to a chlorophyll free version (alfalfa free), as illustrated in the below figure.

Reagents

We offer the D-luciferin at very low prices, see pricelist and book it under service booking.

There are several commercial suppliers of dyes, tracers and labeled cell lines for in vivo imaging, among these:

CT contrast agents can be purchased from Miltenyi Biotec

Mice

B6 albino mice is an advantage to use for work with fluorescence probe, they can be purchased from Taconic farms
Luciferase active mouse models can be acquired from Taconic
