CytoFLEX S (4 laser)

The CytoFLEX is our newest instrument in the facility; in fact we got two identical instruments in at the same time. FACS LSR-II users should consider  changing to these instruments, which will give you more colors. The instrument has 4 lasers and all measurement is done with avalanche Photodiode (APD) detectors provide unmatched red sensitivity and unique compensation workflow, with a digital system with 7-decade display. The resolution of the instrument is particular good <300 nm particles (488 nm side scatter), <80 nm polystyrene particles (405 nm side scatter), rCV 3%. Since the instrument has different software, training is needed also for experienced users.

Specification

Lasers

  • 488 nm solid state laser (50mW)
  • 561 nm solid state laser (30mW)
  • 638 nm solid state laser (50mW)
  • 405 nm solid state laser (80mW)

Optical system

  • Detection of 13 FL-parameters
  • Detection of forward (FSC), side scatter (SSC), and fluorescence light in photo diodes

Blue laser detection (488 nm)*

  • 525/40 nm FITC
  • 690/50 nm PerCP-Cy5.5.

Yellow/green laser detection (561 nm)*

  • 585/42 nm PE
  • 610/20 nm PE-Texas Red or PE-CF594
  • 690/50 nm PE-Cy5.5
  • 780/60 nm PE-Cy7

Red laser detection (638 nm)*

  • 660/10 nm APC
  • 712/25 nm Alexa Flour 700
  • 780/60 nm APC-Cy7

Violet laser detection (405 nm)*

  • 525/40 nm BV480 and BV510
  • 610/20 nm BV605
  • 660/10 nm BV650

We do have additional band pass filters (710/50 and 780/60) that, on request, can be used in the instrument.  This would allow BV colors of longer wavelength among others.

* Each color is just an example, you should ensure that you are using the right color by checking the filter setting.

High Throughput Option

  • The instrument is equipped with a plate loader and switch so you easily can change to single tube loading with 5 ml 12 x 75 mm or 1.2 mL tubes.

Compensations

  • Novel Compensation Library: store fluorescent spillover values of dyes to easily determine the correct compensation matrix with new gain settings
  • Absolute linear gain amplification enables the use of compensation settings between experiments and sample types
